槲皮素是一種重要的膳食類黃酮，在蔬菜、水果、種子、堅果、茶葉和紅酒中均有分布。它在腫瘤預防方面具有各種生物學功能。槲皮素是PI3K、NF-κB和其他參與細胞內信號轉導的激酶的強力抑制劑。它也能輕微抑制Akt1/2，并輕微影響PKC、p38和ERK1/2等 [1]。
許多報告已經證明，槲皮素在癌細胞中具有抗炎、抗氧化和促凋亡等功能，并通過不同的機制抑制腫瘤進展。槲皮素提高了細胞質中Ca2+水平并破壞了線粒體膜電位(MMP)，從而促進了細胞質中細胞色素c的釋放，激活多種半胱氨酸蛋白酶如半胱氨酸蛋白酶-3、-8和-9，并隨后通過線粒體途徑誘導細胞凋亡[2]。槲皮素誘導細胞周期停滯和凋亡，與p53的功能有關。槲皮素可以促進p53磷酸化，因此在mRNA和蛋白質水平上穩(wěn)定p53 [3]。然而，一些研究表明，p53功能受損的細胞對槲皮素介導的細胞毒性和凋亡更為敏感[4]。
體內研究表明，在癌變發(fā)生前期給予槲皮素，能顯著降低小鼠模型中各種化學物質引起的腫瘤負擔，包括苯并(a)芘誘導的肺腫瘤負擔、氧化甲基異腺嘌呤誘導的大鼠結腸癌前病變和N-亞硝基二乙胺誘導的肝癌等[5]。
