MagniSort? 人 CD4 2 Step富集試劑盒設(shè)計用于在第一次消耗單核細(xì)胞后通過陽性篩選對 CD4+ T 細(xì)胞進(jìn)行磁性分離。它已經(jīng)過優(yōu)化，可使用生物素化抗人 CD14 和抗人 CD4 抗體以及鏈霉素親和素包被磁珠從人外周血單核細(xì)胞中分離 CD4+ T 細(xì)胞。在第一步中，單核細(xì)胞被抗人 CD14 抗體結(jié)合，然后被磁珠結(jié)合。當(dāng)置于磁場中時，單核細(xì)胞固定在磁鐵中，而剩余的細(xì)胞則被倒出，用于第二步。第二步，消耗單核細(xì)胞的細(xì)胞被抗人 CD4 抗體結(jié)合，然后被磁珠結(jié)合?，F(xiàn)在，當(dāng)置于磁場中時，CD4+ T 細(xì)胞固定在磁鐵中，而不需要的細(xì)胞則被倒出。

富集后，可通過用抗人 CD4、克隆 OKT4 或 SK3 染色來驗證所選細(xì)胞的純度。
